SEE ALSO...

PDGF-A Antibody (E-10) is a mouse monoclonal IgG2b kappa light chain antibody that detects PDGF-A protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. PDGF-A monoclonal antibody (E-10) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. PDGF-A protein plays a crucial role as a mitogen for mesenchyme- and glia-derived cells, consisting of two disulfide-bonded polypeptide chains, A and B, and existing as three isoforms: PDGF AA, PDGF AB, and PDGF BB. These isoforms exhibit varying affinities for two receptor types, α and β, which are structurally related and possess protein-tyrosine kinase domains. Binding of PDGF-A to receptors induces receptor dimerization and subsequent activation of receptor kinases, with A subunit specifically binding to α receptors with high affinity, while B subunit can interact with both α and β receptors. PDGF-A has been implicated in neurotrophic functions, as evidenced by expression of receptors in oligodendrocyte progenitor cells, suggesting that PDGF-A may regulate growth and differentiation of distinct cell types in developing central nervous system through paracrine and autocrine signaling pathways.

At what molecular weight should the band be seen in Western Blot with this antibody?

Asked by: TinTin
This antibody is reactive with both the precursor and mature form of PDGF-A. On Western blot, you may see it in different forms: as a monomeric A chain or as a dimer, or both.
