PDGF-A Antibody (A-1) is a mouse monoclonal IgG1 antibody that detects PDGF-A in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). PDGF-A plays a crucial role as a mitogen for mesenchyme- and glia-derived cells, consisting of two disulfide-bonded polypeptide chains, A and B, and existing in three isoforms: PDGF AA, PDGF AB, and PDGF BB. These isoforms exhibit varying affinities for two receptor types, α and β, which are structurally related and possess protein-tyrosine kinase domains. The binding of PDGF-A to its receptors induces receptor dimerization and subsequent activation of the receptor kinases, with the A subunit specifically binding to α receptors with high affinity, while the B subunit can interact with both α and β receptors. Notably, PDGF-A has been implicated as a neurotrophic factor, with evidence indicating that its receptors are expressed in oligodendrocyte progenitor cells, while PDGF-B receptors are found on neurons. This suggests that the distinct isoforms of PDGF may play a pivotal role in regulating the growth and differentiation of various cell types in the developing central nervous system through paracrine and autocrine signaling pathways. Anti-PDGF-A antibody (A-1) is an essential tool for researchers investigating the complex roles of PDGF-A in cellular processes and its implications in neurodevelopment.

At what molecular weight should the band be seen in Western Blot with this antibody?

This antibody is reactive with both the precursor and mature form of PDGF-A. On Western blot, you may see it in different forms: as a monomeric A chain or as a dimer, or both.
