paxillin Antibody (B-2) is a mouse monoclonal IgG2b kappa light chain antibody specific for an epitope mapping between amino acids 17-47 near the N-terminus of human paxillin. Paxillin, also known as PXN, is a focal adhesion phosphoprotein localized to the cytoskeleton, playing a pivotal role in cell adhesion, migration, and signal transduction pathways. Paxillin interacts with a variety of proteins, including focal adhesion kinase (FAK), Src family kinases, vinculin, and integrins, serving as a scaffold that assembles multiprotein complexes at focal adhesion sites. These interactions are crucial for transmitting signals from the extracellular matrix to the intracellular environment, influencing processes like cytoskeletal reorganization and cell motility. Phosphorylation of paxillin occurs in response to stimuli such as platelet-derived growth factor (PDGF) treatment and v-src transformation, with kinases like FAK and PYK2 phosphorylating specific tyrosine residues, notably Tyr118. During mitosis, paxillin levels decrease due to proteolytic downregulation, and paxillin undergoes alternative phosphorylation on serine residues, highlighting a role in cell cycle regulation. anti-paxillin antibody (B-2) detects the α, β, and γ isoforms of paxillin from mouse, rat, human, equine, and canine origins in applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates, paxillin (B-2) antibody is a valuable tool for researchers studying the interactions and functions of paxillin in cellular signaling pathways across different species.
