Parkin Antibody (H-8) is a mouse monoclonal IgG2a kappa light chain antibody targeting human Parkin, also referred to as PRKN or parkin RBR E3 ubiquitin protein ligase, and is validated for applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Parkin is a 465-amino acid E3 ubiquitin ligase encoded by the PARK2 gene, which plays a pivotal role in tagging proteins for degradation, a process essential for maintaining cellular homeostasis. Structurally, Parkin contains an N-terminal ubiquitin-like domain and several zinc-finger motifs, including RING (Really Interesting New Gene) domains, which are critical for its E3 ligase activity. These domains facilitate interactions with substrate proteins and the ubiquitin-conjugating machinery, enabling Parkin to regulate protein turnover effectively. The integrity of these structural motifs is vital for Parkin′s function, as mutations within these regions often disrupt its activity and are linked to autosomal recessive juvenile Parkinson′s disease. anti-Parkin antibody (H-8) was generated against amino acids 61-360 of human Parkin, mapping within an internal region of the protein. Parkin is predominantly cytoplasmic but translocates to the mitochondria in response to cellular stress, initiating mitophagy to remove damaged mitochondria. Parkin monoclonal antibody (H-8) is a valuable reagent for studying Parkin′s structural features and their implications in cellular pathways and neurodegenerative diseases.
