pan 14-3-3 Antibody (B-8) is a mouse monoclonal IgG1 kappa light chain antibody that detects the pan 14-3-3 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). pan 14-3-3 (B-8) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. 14-3-3 proteins play a crucial role in regulating numerous cellular processes, particularly those associated with cancer biology, such as apoptosis, mitogenic signaling, and cell-cycle checkpoints. This protein family consists of seven isoforms—14-3-3 β, γ, ε, ζ, η, θ, and Σ—that form dimers, providing two binding sites for ligand proteins. This unique structural feature allows 14-3-3 proteins to facilitate interactions between proteins that might not otherwise associate, which is essential for maintaining cellular homeostasis. The ligands that interact with 14-3-3 proteins typically share a consensus binding motif and are often phosphorylated on serine or threonine residues. By regulating these interactions through mechanisms such as cytoplasmic sequestration, modulation of interaction domains, and prevention of degradation, 14-3-3 proteins significantly influence various signaling pathways. Loss of 14-3-3 expression can lead to a range of pathogenic cellular activities, underscoring 14-3-3′s importance in cellular regulation and disease progression.
