SEE ALSO...

pan 14-3-3 Antibody (B-11) is a mouse monoclonal IgG1 kappa light chain antibody that detects pan 14-3-3 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). pan 14-3-3 (B-11) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. 14-3-3 proteins play a crucial role in regulating numerous cellular processes, particularly those associated with cancer biology, such as apoptosis, mitogenic signaling, and cell-cycle checkpoints. 14-3-3 proteins are located primarily in the cytoplasm and nucleus, where 14-3-3 proteins interact with a wide range of target proteins, influencing their activity and stability. 14-3-3 proteins′ ability to sequester ligands in the cytoplasm is particularly important, as this sequestration prevents ligand degradation and modulates interactions with other cellular components. Such dynamic regulation is essential for maintaining cellular homeostasis and ensuring proper cellular responses to various stimuli. Loss of 14-3-3 expression can lead to dysregulation of these processes, contributing to the development of various diseases, including cancer.

Is this pan antibody for all 14-3-3 isoforms or specific antibody for  14-3-3 beta ? The word (pan) makes me confused.

Asked by: kidarime
Thank you for the question. This antibody is not specific for the beta isoform of 14-3-3 but it will react with different forms of the 14-3-3.

I am using pan 14-3-3 (B-11): sc-133232 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Asked by: Randy McDonald
We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
