SEE ALSO...

Paip1 Antibody (E-7) is a mouse monoclonal IgG2b kappa light chain antibody that detects Paip1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). anti-Paip1 antibody (E-7) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Paip1, known as PABP-interacting protein, plays a crucial role in translation regulation by binding to polyadenylate-binding protein (PABP). This interaction enhances mRNA stability and translation efficiency, which is vital for proper gene expression and cellular function. Paip1 competes with Paip2 for binding to PABP; while Paip1 stimulates translation, Paip2 acts as a repressor by decreasing PABP′s affinity for polyadenylate RNA. The human Paip1 gene encodes a 480 amino acid protein featuring an eIF4A-binding region and a proline-rich N terminus, which are essential for linking mRNA termini and facilitating translation initiation. Paip1 overexpression in COS-7 cells stimulates translation, highlighting its importance in protein synthesis regulation.

Thank you for your suggestion! Based on viewing of the human protein atlas the salivary gland and thyroid gland express the PAIP1 protein! So can either of the two tissues be used as a positive control? Also do endothelial cells show positivity for PAIP1?

I want to use this antibody for IHC, can you please suggest the positive and negative controls to be used with this antibody for validation?

Asked by: Neeti
Thank you for your question. We recommend using human adrenal gland tissue as a positive control for Paip1 with this antibody. We have not characterized negative control tissues for Paip1. According to the Human Protein Atlas (HPA) PAIP1 has RNA detected in all tissues tested with low tissue specificity. Please refer to the link below for this reference. https://www.proteinatlas.org/ENSG00000172239-PAIP1/tissue

Can Paip1 (E-7): sc-365687 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: jenniferc
Thank you for your question. Yes, Paip1 (E-7): sc-365687 has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
