p47phox Antibody (D-10) is a mouse monoclonal IgG1 kappa light chain antibody that detects p47phox of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-p47phox antibody (D-10) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. p47phox plays a crucial role in the immune response, as p47phox is essential for the activation of the NADPH oxidase complex in neutrophils and other phagocytic cells, which generates reactive oxygen species necessary for microbial killing. Mutations in p47phox have been linked to hereditary chronic granulomatous disease, a condition that impairs the body′s ability to fight infections. p47phox is located in the cytosol and translocates to the plasma membrane upon activation, where p47phox interacts with cytochrome b558 and the small G protein Rac to form the functional NADPH oxidase complex. This translocation is vital for the enzyme′s activity, as this process allows for the assembly of the components necessary for superoxide production. Additionally, p47phox contains two Src homology 3 (SH3) domains, and p47phox C-terminal SH3 domain has been shown to interact with the proline-rich domain of p67-phox, indicating that p47phox may facilitate the transport of p67-phox to the membrane, further underscoring p47phox importance in the immune response.
