p-Rb Antibody (F-4) is a mouse monoclonal IgG2a kappa light chain antibody that detects Ser 608 phosphorylated Rb of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). p-Rb (F-4) antibody is available in non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Retinoblastoma protein (Rb) plays a crucial role in regulating cell cycle, particularly at G1-S transition, by acting as a transcriptional repressor. Rb′s ability to control cell proliferation prevents uncontrolled cell growth, which can lead to cancer. Rb achieves this by binding to E2F transcription factors, inhibiting their activity, and preventing expression of genes necessary for S phase entry. Phosphorylation of Rb at specific serine residues, such as Ser 795, during G1 phase leads to inactivation, allowing cell cycle progression. This phosphorylation serves as a key regulatory mechanism and highlights the importance of post-translational modifications in Rb′s functional dynamics, as modifications dictate interactions with other proteins and role in tumor suppression. Understanding Rb regulation through phosphorylation remains essential for developing targeted therapies in cancers, such as retinoblastoma, where Rb gene mutations disrupt normal function.
