p-IκB-α Antibody (B-9) is a mouse monoclonal IgG2b kappa light chain antibody that detects IκB-α phosphorylated at Ser 32 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. p-IκB-α (B-9) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. IκB-α plays a crucial role in regulating the NF-κB signaling pathway, which controls immune responses, inflammation, and cell survival. By sequestering NF-κB dimers in the cytoplasm, IκB-α prevents their translocation to the nucleus, thereby inhibiting the transcription of target genes involved in these processes. Phosphorylation of IκB-α at Ser 32 marks IκB-α for degradation, allowing NF-κB to enter the nucleus and activate gene expression. Understanding IκB-α phosphorylation dynamics is vital for studying various diseases, including cancer and autoimmune disorders, where NF-κB signaling is often dysregulated.

How does addition of HRP to primary Ab affect the WB protocol? Other words what is difference between SC-8404 and SC-8404 HRP? Thank you!

Thank you for your question. The difference between sc-8404 and sc-8404 HRP is that the latter is the primary antibody directly conjugated to horseradish peroxidase (HRP). If using a primary antibody conjugated to HRP, then it is not necessary to use an HRP conjugated secondary antibody for detection in Western blot. Instead, you can incubate the membrane with HRP conjugated primary antibody, and then proceed directly to incubating the membrane in Chemiluminescence Luminol Reagent (sc-2048) and visualization using your standard protocols. Can I use this mouse monoclonal antibody for immunohistochemical staining of mouse tissue?

Yes. We recommend using a directly conjugated mouse monoclonal primary antibody. This mouse monoclonal antibody for p-IκB-α is currently available conjugated to agarose (sc-8404 AC), 500 µg/0.25 ml agarose in 1 ml, for IP; HRP (sc-8404 HRP), 200 µg/ml, for WB, IHC(P) and ELISA; phycoerythrin (sc-8404 PE), fluorescein (sc-8404 FITC), Alexa Fluor® 488 (sc-8404 AF488) or Alexa Fluor® 647 (sc-8404 AF647), 200 µg/ml, for IF, IHC(P) and FCM.

Rated 1 out of 5 by from Showing much larger band size than predictedI bought this antibody 3 weeks ago, I cannot detect the western blot band at the predicted size (40-45 kDa) even when I used positive control, but I got the much larger protein band around 80-90 kDa instead. I used 3% BSA in TBST for blocking and washed for 10 minutes 3 times. It seems like some people from the old comments also have this problem. Can anybody help?

Rated 5 out of 5 by from Very good Ab!The sc-8404 antibody was tested (1: 200, ON 4°) on samples extracted from rat hippocampus using a 12% acrylamide gel. The antibody worked very well. In figure (from left to right): 30ug, 60ug, 80ug and 100ug [marker (from the bottom): 46,57 kDa]

Rated 2 out of 5 by from week signal and additional bandI have used this antibody on mouse brain lysate and I get a very week although detectable signal. However I am struggeling with an additional band around 65-70 kDa. Infact this higher band shows greater response to ischemia than the specific band at the right size, which is quite surprising.
