p-ERK 1/2 Antibody (12D4) is a mouse monoclonal IgG1 antibody that detects p-ERK 1/2 in mouse, rat, human, and canine samples through western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF) applications. p-ERK 1/2 monoclonal antibody (12D4) is available in a non-conjugated form, specifically designed to target the dually phosphorylated forms of ERK 1 and ERK 2 at Thr 202 and Tyr 204. The ERK proteins, which are part of the mitogen-activated protein kinase (MAPK) signaling pathway, play a crucial role in mediating cellular responses to growth factors, hormones, and neurotransmitters. The importance of p-ERK 1/2′s structure lies in its dual phosphorylation mechanism, which is essential for activation. How many amino acids in length is the antigen phosphopeptide of this antibody?

Does this antibody (sc-81492) recognize both dually phosphorylated ERK 1 (thr202 and tyr204) and ERK 2 (thr185 and tyr187)? I have seen only one band in the datasheet.

Thank you for your question. Yes, this antibody, p-ERK 1/2 (12D4): sc-81492 is recommended for detection of Thr 202 and Tyr 204 dually phosphorylated ERK 1 and ERK 2 of mouse, rat, human and canine origin by WB, IP and IF. It is non cross-reactive with non-phosphorylated ERK 1/2, and requires phosphorylation at both the Thr and Tyr sites for reactivity. ERK1 and ERK 2 are very similar in size, and we do not typically observe a doublet or more than one band when using this antibody in Western blotting.
