p-c-Myc Antibody (C-3) is a mouse monoclonal IgG1 antibody raised against amino acids 67-83 of human c-Myc, a crucial transcription factor whose activity is tightly regulated by phosphorylation. p-c-Myc mouse monoclonal antibody (C-3) specifically recognizes c-Myc when phosphorylated at Thr58 or Ser62 and demonstrates broad species reactivity. Anti-p-c-Myc monoclonal antibody (C-3) has been validated for Western blotting, immunoprecipitation, immunofluorescence, immunohistochemistry on paraffin-embedded tissues, and enzyme-linked immunosorbent assay applications. The phosphorylation status of c-Myc at Thr58 and Ser62 serves as a molecular switch controlling protein stability and activity, with each site playing distinct regulatory roles. Ser62 phosphorylation typically occurs first and promotes c-Myc stability and transcriptional activity, while subsequent phosphorylation at Thr58 triggers protein degradation through the ubiquitin-proteasome pathway. This sequential phosphorylation represents a crucial regulatory mechanism, as Ser62 phosphorylation is required for subsequent Thr58 phosphorylation. The interplay between these phosphorylation sites is particularly significant in cancer biology, as mutations affecting Thr58 are frequently found in Burkitt′s lymphoma, leading to increased c-Myc stability and oncogenic activity. ERK kinases typically mediate Ser62 phosphorylation in response to growth signals, while glycogen synthase kinase-3 (GSK-3) phosphorylates Thr58 following priming by Ser62 phosphorylation. This coordinated phosphorylation cascade integrates multiple signaling pathways to fine-tune c-Myc′s transcriptional activity and protein stability. p-c-Myc (C-3) antibody serves as an essential tool for investigating these regulatory mechanisms and their dysregulation in cancer, allowing researchers to monitor the phosphorylation state of c-Myc in various cellular contexts and disease states.
