p-B-Myb Antibody (H-3) is a mouse monoclonal IgG2b kappa light chain antibody that detects Thr 490 phosphorylated B-Myb in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-p-B-Myb antibody (H-3) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. B-Myb, a member of the myb proto-oncogene family, functions as a crucial transcription factor that regulates the cell cycle, particularly facilitating the transition from G1 to S phase. B-Myb is predominantly located in the nucleus, where phosphorylation occurs at specific sites, including threonine 490, by the cyclin A/Cdk2 complex at the onset of S phase. This phosphorylation enhances B-Myb transcriptional activity, thereby promoting cellular proliferation. Notably, phosphorylation of C-terminal residues, such as threonine 447, threonine 490, threonine 497, and serine 581, is essential for B-Myb hyperphosphorylation and subsequent transactivation properties. Additionally, B-Myb interacts with poly(ADP-ribose) polymerase (PARP), which acts as a co-factor that not only binds to B-Myb but also facilitates phosphorylation by the Cyclin/Cdk2 complex, further underscoring the importance of these post-translational modifications in regulating B-Myb′s role in cell cycle progression and transcriptional regulation.
