Ob Antibody (H-5) is a mouse monoclonal IgG1 kappa light chain antibody that detects Ob (leptin) in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-Ob antibody (H-5) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Leptin, the product of the Ob gene, plays a crucial role in regulating energy balance by inhibiting hunger, which in turn helps to manage body weight. Leptin is primarily secreted by adipose tissue and acts on the hypothalamus to signal satiety, making leptin a key player in the complex network of appetite regulation and energy homeostasis. The significance of leptin extends beyond mere appetite control; leptin also influences various physiological processes, including metabolism and immune function. Notably, mutations in the Ob gene can lead to severe obesity and metabolic disorders, underscoring the importance of leptin in maintaining healthy body weight and metabolic health. The cloning and sequence analysis of the mouse Ob gene and its human counterpart have revealed a highly conserved 167 amino acid open reading frame, with an amino acid sequence that is 84% identical between species, highlighting its evolutionary significance. A specific nonsense mutation in codon 105 has been identified in the original congenic C57BL/6J Ob/Ob mouse strain, which serves as a valuable model for studying obesity and related metabolic diseases.
