NUMB Antibody (48) is a mouse monoclonal IgG1 kappa light chain antibody that detects NUMB protein of mouse, rat, human, and canine origin by western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). Anti-NUMB antibody (48) is available in both non-conjugated and various conjugated forms, including agarose and horseradish peroxidase (HRP). NUMB protein plays a crucial role in neuronal cell fate decisions, acting as a signaling adapter with two distinct protein-protein interaction domains: a phosphotyrosine-binding domain and a proline-rich SH3-binding region. This structural configuration is essential for NUMB′s function in mediating interactions with other proteins, such as EPS15, LNX1, and NOTCH1, which are vital for the regulation of cell fate during development. In mammals, NUMB homologs are implicated in determining cell fates, and studies have shown that conditional mouse mutants lacking NUMB in developing sensory ganglia exhibit reduced axonal arborization in afferent fibers, highlighting NUMB′s importance in neural development. Additionally, alterations in cellular calcium homeostasis can influence NUMB-dependent cell fate decisions, further underscoring NUMB′s significance in the nervous system′s development. In chicken, NUMB (c-NUMB) is localized to the basal cortex of mitotic neuroepithelial cells, indicating NUMB′s role in cellular organization during neurogenesis.

When should the TransCruz reagent sc-136554 X be used instead of the standard form sc-136554?

Thank you for your question. We recommend using the TransCruz reagent form when performing ChIP or GelShift assays. This form can also be used in other applications such as Western Blot or IHC, but the dilution should be adjusted by a factor of 10 to account for the higher original concentration.
