Npl4 Antibody (D-1) is a mouse monoclonal IgG1 kappa light chain antibody that detects Npl4 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-Npl4 antibody (D-1) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Npl4, consisting of 608 amino acids, is primarily located in the endoplasmic reticulum and nuclear membrane, where Npl4 plays a crucial role in the cellular quality control system. By forming a complex with p97/Cdc48p and Ufd1p, Npl4 is essential for recognizing and processing ubiquitinated inositol 1,4,5-trisphosphate receptors within the endoplasmic reticulum, facilitating their delivery to the proteasome for degradation. This function is vital for maintaining cellular homeostasis and regulating intracellular signaling pathways. Additionally, Npl4 features multiple post-translational modifications, including potential N-myristoylation and N-glycosylation sites, as well as phosphorylation sites, which may influence stability and interactions with other proteins. Notably, mutations in the NPL4 gene can lead to significant defects in nuclear envelope morphology and impairments in nuclear protein import and RNA export, underscoring Npl4′s importance in cellular function and integrity.
