Npl4 Inhibitors

Npl4 inhibitors are primarily focused on the disruption of the p97/VCP complex, of which Npl4 is an integral part. The chemical inhibitors listed target the ATPase activity of p97, which is essential for the function of the p97-VCP-UFD1-NPL4 complex in cellular processes such as the endoplasmic reticulum-associated degradation (ERAD) pathway, and the overall maintenance of protein homeostasis within the cell. These inhibitors are diverse in structure and mode of action but converge on the common goal of impairing the ATPase activity of p97.

The inhibition of p97's ATPase activity results in the malfunctioning of the p97-VCP-UFD1-NPL4 complex, leading to the accumulation of polyubiquitinated proteins, which can interfere with various cellular processes. Inhibitors such as CB-5083, NMS-873, and DBeQ bind to distinct sites on p97, either at the allosteric or ATP-binding sites, to exert their inhibitory effects. By destabilizing the proper functioning of this complex, these compounds indirectly inhibit the activity of Npl4.
