Neurocan is an essential component of the extracellular matrix in the central nervous system, where neurocan interacts with various matrix components such as heparin, Tenascin-R, HB-GAM, and NCAM, indicating involvement in axon guidance and neurite outgrowth. Neurocan consists of 1,257 amino acids and undergoes proteolytic processing in the adult central nervous system, resulting in an N-terminal fragment that localizes to the cytoplasm of glial cells. Notably, neurocan expression, including the re-expression of the neonatal isoform, significantly increases in adults within the glial scar following cortical injury, underscoring a potential role in response to neural damage and repair mechanisms.
