Neu Antibody (A-2) is a mouse monoclonal IgG1 kappa light chain antibody that detects Neu in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). anti-Neu antibody (A-2) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Neu, also known as ErbB-2 or HER2, is a member of the epidermal growth factor receptor (EGFR) family, which consists of receptor tyrosine kinases that are often overexpressed in numerous carcinomas. Upon ligand binding, Neu can form homodimers or heterodimers with other EGFR family members, such as ErbB-3. This heterodimerization is crucial as it leads to the recruitment of heregulin, which activates phosphoinositide 3-kinase, a key player in various cellular processes including cell growth, survival, and motility. The aberrant expression of Neu, which occurs in 20-30% of breast cancers, is associated with poor prognosis, making Neu a vital biomarker for disease progression and a promising target for therapeutic interventions. Neu monoclonal antibody (A-2) enables researchers to gain insights into Neu′s role in cancer biology and explore potential treatment strategies.

What application is the blocking peptide sc-393712 P appropriate for?

Asked by: Dr Ninau Qelp
Thank you for your question. The blocking peptide is intended for use as a negative control, by pre-adsorbing the mouse monoclonal antibody against the antigen. For full protocol details, please contact our Technical Services Department or view our online protocol here: https://www.scbt.com/scbt/resources/protocols/peptide-neutralization

Rated 1 out of 5 by from did not work for immunofluorescence (pig)Immunohistochemistry: the antibody was tested on mammary parenchyma and small intestine from 6 months old gilts. Antigen retrieval was performed in citrate buffer, 30'. Primary antibody: 1:50 dilution, 1h, RT. Secondary antibody: Alexa Fluor 488 (1:500), 1h, RT. Results: negative; nonspecific signal was observed both in epithelial and connective fractions. Immunocytochemistry: the antibody was tested on porcine mammary epithelial cells isolated from 6 months old gilts. Cells were fixed with acetone-methanol (1:1) and stained as above (see immunohistochemistry). Primary antibody was diluted 1:100. Result: negative.
