NEPH1 Antibody (F-6) is a mouse monoclonal IgG2a kappa light chain antibody that detects NEPH1 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). NEPH1 Antibody (F-6) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. NEPH1, a member of the immunoglobulin superfamily, is primarily located in the glomerular visceral epithelial cells, or podocytes, which are essential for maintaining the selective filtration barrier of the renal glomerulus. The proper localization of NEPH1 is crucial as NEPH1 interacts with nephrin and ZO-1 at the glomerular slit diaphragm, forming a complex that is vital for kidney function. Disruption of NEPH1 expression, similar to nephrin, can lead to severe conditions such as nephrotic syndromes and proteinuria, ultimately resulting in perinatal death. The interaction between NEPH1 and nephrin occurs through the extracellular domain, while binding to ZO-1 is mediated by a PDZ binding motif in the cytoplasmic tail. A putative threonine phosphorylation site within the cytoplasmic domain has been identified, and mutation disrupts the interaction with ZO-1, suggesting that phosphorylation may play a regulatory role in the assembly of this critical signaling complex.
