NCK2 Antibody (8.8) is a mouse monoclonal IgG2b kappa light chain antibody that detects NCK2 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. Anti-NCK2 antibody (8.8) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. NCK2 plays a crucial role in cellular signaling as a member of the NCK family of SH2/SH3 adaptor proteins, which also includes NCK1. These proteins are essential for coupling tyrosine kinase signaling pathways, such as those activated by epidermal growth factor (EGF) and platelet-derived growth factor (PDGF), to downstream signaling proteins. NCK2 binds to human PDGFR-β at tyrosine 1009, and overexpression has been shown to inhibit PDGF-induced membrane ruffling and lamellipod formation, thereby regulating cell motility and morphology. Additionally, the SH2 domain of NCK2 has been implicated in the inhibition of EGF- and PDGF-induced DNA synthesis, highlighting a role in controlling cell proliferation. The interaction of NCK2 with proline-rich domains on proteins like PAK, a known regulator of the actin cytoskeleton, further emphasizes importance in mediating cellular responses to growth factors.
