N-SMase2 Antibody (G-6) is a mouse monoclonal IgG2a kappa light chain antibody that detects N-SMase2 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). N-SMase2 (G-6) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. N-SMase2, also known as NSMASE2 or sphingomyelin phosphodiesterase 3 (SMPD3), is a 655 amino acid enzyme that plays a crucial role in lipid metabolism by catalyzing the hydrolysis of sphingomyelin into ceramide and phosphocholine. This reaction is vital for regulating cell growth and apoptosis, making N-SMase2 an important player in cellular signaling pathways. N-SMase2 is primarily localized to the membrane of the Golgi apparatus, where activation occurs by unsaturated fatty acids and phosphatidylserine, linking N-SMase2 to processes such as growth suppression and postnatal development. Notably, N-SMase2 expression increases during the G0/G1 phases of the cell cycle, with optimal enzymatic activity at a slightly basic pH of 7.5. Deficiencies in N-SMase2 have been associated with chondrodysplasia, a genetic disorder that results in impaired bone growth, leading to short stature and underdeveloped joints, highlighting N-SMase2′s significance in skeletal development and overall health.

To Santa Cruz, I have purchased the antibody for SMPD3 (N-SMase2, G-6, sc-166637), but there are so many bands. It is not clear which band is the one indeed corresponding to that of SMPD3. Could you give a suggestion?

Long story short, don't be fooled, while this Ab gives a clean band around 70kDa, this couldn't be verified by siRNA knockdown of nSMase2. The discontinued Rabbit polyclonal Ab was better and was verified perfectly by siRNA knockdown. Don't buy!
