N/R-cadherin Antibody (H-4) is a mouse monoclonal IgG2b kappa light chain antibody designed to detect both N-cadherin and R-cadherin proteins across multiple species, including mouse, rat, human, avian, equine, canine, and bovine, utilizing applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). N/R-cadherin monoclonal antibody (H-4) targets a precise epitope within amino acids 808-837 of the C-terminal cytoplasmic domain of human N-cadherin, ensuring accurate and reliable binding. N/R-cadherin proteins are crucial members of the cadherin family, which are calcium-dependent adhesion molecules essential for mediating cell-cell adhesion, maintaining tissue structure, and facilitating morphogenesis. Located primarily at the plasma membrane, N/R-cadherin plays a pivotal role in the formation and maintenance of adherens junctions, which are vital for cellular communication and the structural integrity of tissues. The structure of N/R-cadherin comprises multiple extracellular cadherin repeats that mediate homophilic binding between cells, a single transmembrane domain that anchors the protein to the cell membrane, and a relatively short cytoplasmic domain that interacts with various intracellular proteins. These structural features are critical for the protein′s function in cell adhesion and signaling. Post-translational modifications of N/R-cadherin, such as phosphorylation, are important as they regulate protein interactions and stability, influencing processes like cell migration, differentiation, and tissue remodeling. N/R-cadherin interacts with several key proteins, including beta-catenin, which links cadherins to the actin cytoskeleton, α-catenin, which connects beta-catenin to actin filaments, and p120 catenin, which regulates cadherin stability and turnover at the cell membrane. These interactions are essential for the dynamic regulation of cell adhesion and the maintenance of cellular architecture. Anti-N/R-cadherin antibody (H-4) is available in both non-conjugated form and a variety of conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. N/R-cadherin (H-4) antibody is instrumental in research areas such as developmental biology, cancer metastasis, and neurobiology, where understanding the precise mechanisms of cell adhesion and signaling is fundamental.

This antibody is suggested as a replacement for the discontinued: Sc-7939. It does not work for IF in our system, while Sc-7939 worked extremely well. Do you have another suggested replacement for Sc-7939 for IF protocols?

Can the peptide sc-271386 P be used in Western Blot as a positive control?

Thank you for your question. The blocking peptide sc-271386 P is a short synthetic peptide intended for use as a negative control by preadsorption. It is not appropriate for use as a positive control in Western Blot.

Rated 1 out of 5 by from Did not work for IFI ordered this antibody as it was a recommended replacement for Sc-7939, which was discontinued. We routinely used Sc-7939 for IF staining on early mouse embryos and it worked wonderfully. This antibody does not work for IF in the same system and hence cannot be used as a replacement in our case.
