MyoD Antibody (G-1) is a mouse monoclonal IgG2b kappa light chain antibody specifically designed to detect Myogenic Differentiation 1 (MyoD), also known as MYOD1, a crucial transcription factor within the Myogenic Regulatory Factors (MRFs) family that includes myogenin, Myf5, and MRF4 (herculin). anti-MyoD antibody (G-1) is raised against amino acids 1-318 of mouse MyoD, targeting a region that encompasses both the basic helix-loop-helix (bHLH) domains and other critical regions essential for MyoD′s function. MyoD plays a vital role in muscle cell differentiation by binding to E-box sequences (CANNTG) in DNA, thereby regulating the transcription of genes necessary for muscle development and regeneration. Post-translational modifications of MyoD, such as phosphorylation, acetylation, and ubiquitination, are essential for modulating stability, activity, and interactions with other proteins, making the detection of these modifications important for understanding the regulation of myogenesis. MyoD (G-1) monoclonal antibody is validated for use in western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA) applications across multiple species, including mouse, rat, and human, without cross-reacting with other members of the MyoD family. MyoD (G-1) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates, providing versatility for diverse experimental protocols.

I am using the MyoD antibody (G-1, sc-377460) and need to confirm the exact epitope it recognizes. My protein is a truncated MYOD1 (from Cys101 to C-terminus with an N-terminal His-tag). Could you clarify if this antibody binds to the C-terminal region?

Thank you for your question. MyoD Antibody (G-1): sc-377460 was raised against amino acids 1-318 of MyoD of mouse origin, representing full-length protein. No epitope mapping has been performed for this antibody as of yet, so its exact binding site is not known at this point in time.

Can sc-377460 recognize bovine MyoD in Western blot analysis and IHC?

I tried this antibody (x1000) to detect recombinant bovine MyoD expressed in 293T cells and native MyoD in differentiating C2C12 cells by Western analysis. Approximately 45 kDa protein was clearly detected in 293T cells transfected with MyoD-expressing vector and differentiating C2C12 cells. However, a strong non-specific signal (~50 kDa) with some fainter bands were also detected in both cell line. Therefore, I would like to conclude that this antibody can detect bovine MyoD in the Western blot analysis. M Kobayashi

いつもお世話になります。 御社の下記の抗体を使用しています。 MyoD (G-1)  データシート等に掲載されているWBの予測分子量は約45kDaです。 しかしながら、この45kDaに加え、約70kDa近辺に強いバンドを検出しています。 我々の結果の場合、45kDaより70kDaのバンドの方が強く検出されます。 御社のホームページ上でも70kDa近辺に強いバンドが検出されているデータが掲載されています。 この70kDaのバンドの由来に関する情報をお持ちでしたら、ご教示ください。

I had used MyoD C-20 to detect MyoD in chicken myoblasts. It was great. However, C-20 was discontiuned and MyoD G-1 was produced instead. So, I brought the G-1, but the detection was not as good as C-20. Is there any way to get C-20?

Thank you for your question. Unfortunately, the rabbit polyclonal antibody, MyoD (C-20): sc-304, has been discontinued since 2016. It is no longer available for sale or distribution. We do not have any data or product citations yet using this mouse monoclonal antibody in chicken or avian samples. Please contact our Technical Service Department and we could assist you by performing sequence analysis to predict potential cross reactivity of our MyoD antibodies with chicken samples.

Thank you for your question. This antibody hasn’t been tested in bovine samples. The exact epitope recognized by this mouse monoclonal antibody has not yet been determined, so we can not perform sequence analysis at this time to predict potential cross reactivity with that species.

Buenos días, estoy haciendo una inmunoprecipitacion con MyoD y quiero ver su anticuerpo HDAC1, pero la banda que me sale a 60 kDa no tengo claro si es HDAC1 o es la IgG, podrian decirme el peso al que me deberian salir las IgG??
