Mucin 2 Antibody (F-2) is a mouse monoclonal IgG1 kappa light chain antibody that detects the Mucin 2 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-Mucin 2 antibody (F-2) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Mucin 2, also known as oligomeric mucus/gel-forming mucin, plays a crucial role in forming protective mucus layers in the intestines and airways, which is essential for maintaining epithelial integrity and facilitating the clearance of pathogens and debris. Mucin 2′s structure is characterized by a high degree of glycosylation and a unique arrangement of variable number tandem repeats (VNTRs), which contribute to gel-forming properties and ability to trap water, thus providing a viscous barrier that protects underlying tissues. Alterations in Mucin 2 expression and function have been linked to various gastrointestinal disorders, including inflammatory bowel disease and colorectal cancer, highlighting Mucin 2′s significance in both health and disease.

We have been unsuccessful at obtaining Muc2 bands in HT29 MTX E12 lysates (goblet cells) which should be producing high levels. What is your recommendation for the lysis as well as what type of gel, since the molecular weight is potentially 600kD (dimer)?

Thank you for your question. We would recommend using a low percentage gel such as 4-6% for this very large protein. We use RIPA lysis buffer in our lysate preparations, and Electrophoresis Sample Buffer for our Western blot.

What is the size expected when performed a western blot analysis of Mucin 2 expression in mouse?

Appears to be specifically staining the glycosylated forms of Muc2. It never shows perinuclear staining. Clone CCP58 for MUC2 antibodies should show that better.

I ordered the free sample, the unconjugated mouse monoclonal Mucin 2 IgG1 Antibody (F-2) antibody. I tried three different kinds of secondary antibodies: Goat anti mouse IgG (H+L), A21422, Invitrogen, donkey anti mouse IgG (H+L), A21202, Invitrogen, and goat anti mouse IgG1, A21127, Invitrogen.
