MOF Antibody (8C4C4) is a mouse monoclonal IgG1 antibody that detects the MOF protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. Anti-MOF antibody (8C4C4) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The MOF protein plays a crucial role in dosage compensation, a process that ensures equal expression of X-linked genes in males and females. In Drosophila, this process is achieved through the formation of a dosage compensation complex (DCC) that includes proteins such as maleless and male-specific lethal proteins, which work together to enhance transcription from the male X chromosome. MOF is essential for this process as MOF acetylates histone H4 at lysine 16, a modification that facilitates the binding of the DCC to the X chromosome. In mammals, MOF, also known as hMOF, MYST1, or MOZ, is part of the MYST family of histone acetyltransferases, characterized by a unique C2HC-type zinc finger that interacts with nucleosomes and histone substrates. This structural feature is vital for MOF′s enzymatic activity, allowing MOF to regulate gene expression through histone modification, thereby influencing various biological processes, including development and cellular response to environmental stimuli.

I would like to ask if the MOF antibody recognizes the acetylated form in lysine 274, the unmodified form or both indistinctly.

Thank you for your question. This antibody was raised against a recombinant protein corresponding to amino acids 132-283 of MOF (MYST1) of human origin. The exact epitope has not been mapped. The antibody appears to detect MOF in all forms.

Are there any issues with using sc-81163: MOF (8C4C4) mouse monoclonal primary antibody on mouse samples?

To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent,we recommend using a directly conjugated primary antibody.
