MMP-12 Antibody (G-2) is a mouse monoclonal IgG1 kappa light chain antibody that detects MMP-12 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). MMP-12 (G-2) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. MMP-12, also known as macrophage metalloelastase, plays a crucial role in the degradation of elastin, a key component of the extracellular matrix, which is vital for maintaining the structural integrity and elasticity of tissues. MMP-12′s ability to degrade elastin is particularly important in inflammatory disorders, where MMP-12 may facilitate macrophage migration through the epidermal and vascular basement membranes, contributing to tissue remodeling and repair processes. Additionally, MMP-12 gene transcription is differentially regulated by various stimuli such as phorbol ester, lipopolysaccharide, and staphylococcal enterotoxin B, highlighting MMP-12′s involvement in diverse physiological and pathological processes. MMP-12′s enzymatic activity depends on calcium and zinc ions, which are essential for catalytic function, further underscoring MMP-12′s significance in extracellular matrix dynamics and immune responses.

Does this antibody target the pro-form or active form of Mmp12 or both?

Asked by: Zhipeng
Thank you for your question. The zymogen form for MMP-12 is about ~54 kDa, which is activated to the 45-48 kDa form, which further breaks down to a cascade of active forms and resulting in a common 22 kDa form. Which form you will see depends on what form is present in your sample. Our data with this antibody generally shows the active 48kDa form.
