MLH1 Antibody (G168-15) is a mouse monoclonal IgG1 antibody that detects MLH1 in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). MLH1 plays a crucial role in the DNA mismatch repair (MMR) process, which is vital for maintaining genetic stability by correcting DNA replication errors. MLH1 (G168-15) antibody helps identify deficiencies linked to various human cancers, particularly due to mutations or epigenetic silencing of the MLH1 gene. MLH1 is predominantly located in the nucleus, where MLH1 forms a complex with other MMR proteins to identify and repair mismatched DNA bases, thereby preventing mutations that could lead to tumorigenesis. MLH1 expression is notably observed in various tissues, including lymphocytes, heart, colon, breast, lung, spleen, testis, prostate, thyroid, and gall bladder, with MLH1 methylation being a common feature in several ovarian tumors. Loss of MLH1 expression is associated with a mutated phenotype and microsatellite instability, which are significant indicators of cancer predisposition. In hereditary nonpolyposis colorectal cancer (HNPCC), pathogenic mutations in the MLH1 gene highlight MLH1′s importance in cancer risk assessment. Notably, certain cancer cell lines, such as leukemia CCRF-CEM and colon HCT 116, exhibit complete deficiency of MLH1, while MLH1 is expressed in a significant percentage of melanomas and squamous cell carcinomas, underscoring MLH1′s relevance in cancer biology and potential as a therapeutic target.
