MITF Antibody (21D1418) is a mouse monoclonal IgG1 kappa light chain antibody that detects MITF in mouse, rat, and human samples through western blotting (WB) and immunoprecipitation (IP). Anti-MITF antibody (21D1418) is available in a non-conjugated form, allowing for versatile applications in research. The microphthalmia-associated transcription factor (MITF) plays a crucial role in the development and differentiation of melanocytes, which are responsible for pigment production in the skin, hair, and eyes. MITF (21D1418) monoclonal antibody recognizes a protein essential for pigmentation and regulation of various genes involved in melanocyte function, including tyrosinase, TRP1, and TRP2, which are vital for melanin synthesis. The presence of basic helix-loop-helix (HLH) and leucine zipper (LZ) domains in MITF allows for homodimer formation and direct DNA binding, activating gene expression through E-box elements. MITF activity is modulated by post-translational modifications, such as phosphorylation on Ser73 via the MAPK pathway, which enhances interaction with co-activators from the p300/CBP family, thereby increasing transcriptional activity. MITF-deficient mice exhibit a complete absence of melanocytes, highlighting MITF′s indispensable role in melanocyte biology and pigmentation.

I am using MITF (21D1418): sc-52938 antibody for immunoprecipitation, how much antibody should I use?

Thank you for your question. We recommend using 1–10 µl (i.e., 0.2–2 µg) of antibody with 1 ml of cell lysate. However, optimal antibody concentration should be determined by titration.
