    MGMT Antibody (F-5) is a mouse monoclonal IgG2a kappa light chain antibody raised against the full-length human MGMT protein (amino acids 1-207), showing reactivity with MGMT of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). MGMT, also known as O6-methylguanine-DNA methyltransferase or O-6-alkylguanine DNA alkyltransferase (AGT), is a crucial DNA repair enzyme that maintains genomic integrity by removing mutagenic and cytotoxic alkyl groups from the O6 position of guanine in DNA. Structurally, MGMT is a single-chain polypeptide consisting of approximately 207 amino acids, featuring a conserved active site with a critical cysteine residue (Cys145 in humans). MGMT (F-5) antibody directly transfers the alkyl group from the damaged DNA to itself, inactivating the enzyme in a "suicide" reaction, which underscores the importance of its structure for efficient DNA repair. Post-translational modifications of MGMT, such as ubiquitination, are significant as they regulate protein stability and degradation. Ubiquitination targets MGMT for proteasomal degradation, controlling cellular levels and modulating DNA repair capacity of the cell. The regulation of MGMT through these modifications is crucial because overexpression of MGMT can confer resistance to alkylating chemotherapeutic agents, impacting cancer treatment outcomes. Understanding the structure and post-translational modifications of MGMT is essential for cancer research and therapeutic development.
