MGMT Antibody (C-5) is a mouse monoclonal IgG1 kappa light chain antibody raised against the full-length human MGMT protein (amino acids 1-207), showing reactivity with MGMT of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). MGMT, also known as O6-methylguanine-DNA methyltransferase or alkylguanine DNA alkyltransferase (AGT), is a crucial DNA repair enzyme predominantly located in the cell nucleus, where MGMT directly interacts with genomic DNA to remove mutagenic and cytotoxic alkyl groups from the O6 position of guanine. This nuclear localization is essential as MGMT promptly repairs DNA damage before replication, maintaining genomic stability and preventing carcinogenesis. Structurally, MGMT is a single-chain polypeptide consisting of approximately 207 amino acids, featuring a conserved active site with a critical cysteine residue (Cys145 in humans) that accepts the alkyl group from damaged DNA through a direct reversal repair mechanism. This "suicide" inactivation of MGMT after alkyl group transfer underscores the importance of MGMT structure for function in DNA repair. I am using MGMT (C-5): sc-271154 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?
