MerTK Antibody (B-1) is a mouse monoclonal IgG2b kappa light chain antibody that detects MerTK protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-MerTK antibody (B-1) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. MerTK, also known as c-Mer, is a vital member of the Mer/Axl/Tyro3 receptor kinase family and consists of 984 amino acids, featuring one tyrosine kinase domain, two fibronectin type-III domains, and two immunoglobulin-like C2-type domains. MerTK is predominantly located on the surface of macrophages and plays a crucial role in the immune response, particularly in the clearance of apoptotic cells, which is essential for maintaining tissue homeostasis and preventing autoimmune diseases. MerTK′s signaling pathway is activated through interaction with phosphatidylinositol-specific phospholipase C γ2 (PI-PLC γ2), which is critical for the phagocytic process in the retinal pigment epithelium (RPE). Mutations in the MerTK gene can disrupt this pathway, leading to conditions such as autosomal recessive retinitis pigmentosa, characterized by the degeneration of retinal photoreceptor cells. MerTK′s ability to mediate macrophage activation and promote efficient removal of dying cells underscores its importance in both immune regulation and retinal health, making MerTK (B-1) antibody an invaluable tool for researchers studying these processes.

When using this antibody to do IHC, the cellular location is what, nucleus, cytosol or membrane?

Thank you for your question. MerTK is a transmembrane protein that possesses three different domains: Amino acids 19 – 497 represent its extracellular topological domain. Amino acids 498 – 518 correspond to its helical transmembrane domain. Amino acids 519 – 994 form a cytoplasmic topological domain. Since MerTK Antibody (B-1): sc-365499 has been raised against the N-terminal amino acids 1 – 222, the antibody should recognize the extracellular part of MerTK.

Rated 5 out of 5 by from Works great in FACSMerTK used in Flow Cytometry. 1*10^5 PMA-differentiated THP-1 macrophages were were stained with 2.5ul MerTK in 4% FCS/PBS for 30 minutes on ice, washed and incbuated with anti mouse secondary ab conjugated to CF(tm) 640R. Cells were read in a Accuri C6 using the FL4 sensor.

Place the sections into the boil in 600 mL Tris-EDTA buffer for 20 min, and maintained at a room temperature to cool. then were blocked with 10% normal goat serum for 10 min at 37 °C. Rated 4 out of 5 by from Potent and fairly specific antibodyThis monoclonal antibody was independently tested in-house by Kinexus Bioinformatics Corporation and validated for use for Western blotting. There was strong detection of the ~140-205 kDa target proteins in an A431/HepG2/MCF7 cell lysate mix, but the target was not detected in mouse tissues (brain/testes/thymus/muscle mix). No major cross-reactivities were observed at 2 µg/ml antibody concentration with off-target proteins in the mouse tissues tested, but the antibody can generate a slightly higher than typical background when used at 2 µg/ml. A ~52 kDa medium cross-reactive protein was detectable in the A431/HepG2/MCF7 cell lysate mix.
