Mcl-1 Antibody (22) is a mouse monoclonal IgG1 kappa light chain antibody, specifically raised against the full-length human Mcl-1 protein, and is effective in detecting both the long and short forms of Mcl-1 in human samples by western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). Mcl-1, also called Myeloid cell leukemia-1, Mcl1, and Gm52627, is a pivotal member of the Bcl-2 family of proteins, which are critical regulators of apoptosis. Functioning as an anti-apoptotic protein, Mcl-1 promotes cell survival by binding to and inhibiting pro-apoptotic factors such as Bak and Bax, thereby preventing mitochondrial outer membrane permeabilization and the release of cytochrome c. This function is particularly vital in the hematopoietic system, where Mcl-1 is necessary for the maintenance and homeostasis of early progenitor cells. Dysregulation and overexpression of Mcl-1 are implicated in the development and progression of various cancers, including leukemia, lymphoma, and solid tumors, as Mcl-1 allows malignant cells to evade apoptosis and continue proliferating uncontrollably. The stability and activity of Mcl-1 are regulated by several signaling pathways, including the PI3K/Akt pathway and glycogen synthase kinase 3 (GSK3), making Mcl-1 a significant target for therapeutic intervention in cancer treatment.
