Mcl-1 Antibody (22) is a mouse monoclonal IgG1 kappa light chain antibody, specifically raised against the full-length human Mcl-1 protein, and is effective in detecting both the long and short forms of Mcl-1 in human samples by western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). Mcl-1, also called Myeloid cell leukemia-1, Mcl1, and Gm52627, is a pivotal member of the Bcl-2 family of proteins, which are critical regulators of apoptosis. Functioning as an anti-apoptotic protein, Mcl-1 promotes cell survival by binding to and inhibiting pro-apoptotic factors such as Bak and Bax, thereby preventing mitochondrial outer membrane permeabilization and the release of cytochrome c. This function is particularly vital in the hematopoietic system, where Mcl-1 is necessary for the maintenance and homeostasis of early progenitor cells. Dysregulation and overexpression of Mcl-1 are implicated in the development and progression of various cancers, including leukemia, lymphoma, and solid tumors, as Mcl-1 allows malignant cells to evade apoptosis and continue proliferating uncontrollably. The stability and activity of Mcl-1 are regulated by several signaling pathways, including the PI3K/Akt pathway and glycogen synthase kinase 3 (GSK3), making Mcl-1 a significant target for therapeutic intervention in cancer treatment. Understanding and detecting Mcl-1 expression levels are therefore critical in cancer research and drug development. Mcl-1 (22) antibody is available in both unconjugated form and conjugated forms with agarose, horseradish peroxidase, phycoerythrin, fluorescein isothiocyanate, and multiple Alexa Fluor® conjugates, providing researchers with versatile tools for studying Mcl-1 in various experimental applications.

Dear Sir or Madam, I have tried this antibody on a panel of cell lines, one of them Jurkat which should give a strong signal. So far I only get very faint bands and plenty of unspecific ones. Do you have any advice please? ,Thank you

Does this antibody interact with mouse MCL1 or it solely probes for human MCL1?

Can it be used in paraffin embedded tissues for immunohistochemical analysis? As I haven't found something from the citations.

Thank you for your question. We have not yet tested our mouse monoclonal Mcl-1 Antibody (22): sc-12756 for immunohistochemistry on formalin-fixed paraffin-embedded tissue sections. As a result, the application of IHC(P) is not currently recommended. This antibody is extremely well cited, but it is most frequently used for Western blot (WB) and immunoprecipitation (IP). We would recommend an alternative clone for IHC(P), Mcl-1 Antibody (G-7): sc-74437 (see link below). This antibody has been tested in-house and has excellent published data for IHC(P) shown on our website. https://www.scbt.com/scbt/product/mcl-1-antibody-g-7

Rated 1 out of 5 by from It does not workWe ordered this antibody as alternative to rabbit polyclonal we had from the compony. We tried western blot in parallel with the antibody we had before. This monoclonal gave us no specific signal for MCL-1. We also had lysates from cells over expressing MCL-1. The antibody did not work for these lysates too.
