MAP LC3α/β Antibody (G-4) is a mouse monoclonal IgG1 kappa light chain antibody engineered to recognize both MAP LC3α and MAP LC3β, also known as LC3A and LC3B or MAP1LC3A and MAP1LC3B, respectively. Anti-MAP LC3α/β antibody (G-4) targets an internal epitope spanning amino acids 33-56 of the human MAP LC3β protein, enabling its use in various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA) across human, mouse, and rat species. MAP LC3α/β plays a pivotal role in the regulation of autophagy, a critical cellular process for maintaining homeostasis by degrading and recycling damaged organelles and proteins. Post-translational modifications, such as the lipidation of MAP LC3β from LC3-I to LC3-II, are essential for its association with autophagosome membranes, facilitating the formation and maturation of autophagosomal vacuoles. Additionally, MAP LC3α exists in two alternatively spliced isoforms that are expressed in tissues including the testis, brain, heart, liver, and skeletal muscle, but are notably absent in the thymus and peripheral blood leukocytes, highlighting its specialized functions in these tissues. MAP LC3β, present in both cytosolic and membrane-bound forms, is primarily expressed in the heart, testis, brain, and skeletal muscle, underscoring its importance in neuronal development and cellular structural integrity. MAP LC3α/β (G-4) antibody is available in both non-conjugated and various conjugated forms, such as horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® derivatives, providing versatility for diverse experimental setups.

Due to some issues, I will not be able to do methanol fixation for IF and have to stick with 4% PFA followed by TritonX-100 permeabilization. Will this antibody work in those conditions?

Thank you for your question. This antibody, MAP LC3α/β (G-4): sc-398822, has not yet been tested on formalin or PFA fixed tissues or cells so we do not know if it will work in this application. However, we would recommend including a permeabilization step, such as with Triton X-100, if using 4% PFA as a fixative.

What fixation method do you recommend for IF (human fibroblasts)?

We diluted it with 3% milk powder at 1/500 and 1/1000 ratios. And each time we capture the signal in the range of 55-72 kDa. Where are we going wrong?
