Mad2 Antibody (C-3) is a mouse monoclonal IgG1 kappa light chain antibody that detects Mad2 protein of Saccharomyces cerevisiae origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Mad2 (C-3) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Mad2 protein plays a crucial role in the cell cycle by acting as a key regulator of the mitotic spindle assembly checkpoint, which ensures cells do not proceed to anaphase until all chromosomes are properly attached to the spindle apparatus. This function is vital for maintaining genomic stability, as cells with dysfunctional Mad2 can experience improper chromosome segregation, leading to aneuploidy and potential tumorigenesis. Studies have shown that breast carcinoma cell lines with reduced Mad2 expression, such as T47D, can complete mitosis even in the presence of nocodazole, a drug that disrupts spindle assembly, highlighting Mad2′s importance in cell division fidelity. Mad2 localizes to unattached kinetochores during prometaphase and releases upon proper spindle fiber attachment, indicating a dynamic role in monitoring kinetochore-spindle interactions. Human Mad2 has been implicated in insulin receptor signaling, associating specifically with the insulin receptor but not with the insulin-like growth factor receptor, suggesting a broader role in cellular signaling pathways beyond cell cycle function.
