Ly-6D Antibody (F-6) is a mouse monoclonal IgG2b antibody that detects Ly-6D in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Ly-6D, also known as E48 antigen, is a 128 amino acid glycoprotein predominantly expressed in squamous cell carcinoma cell lines and squamous cell epithelial tissues. Ly-6D plays a crucial role in cell adhesion and migration, particularly in head and neck cancers, where upregulation is associated with poor prognosis. Notably, Ly-6D has been implicated in the regulation of fucosylated E-selectin ligands and interacts with protein FX, which is essential for the synthesis of GDP-L-fucose. This interaction suggests that Ly-6D may influence tumor cell adhesion to inflamed vessel walls that express E-selectin, thereby contributing to the metastatic potential of cancer cells. The presence of a signal peptide, along with theoretical phosphorylation and myristoylation sites, further indicates Ly-6D′s involvement in cellular signaling and membrane localization, making Ly-6D a valuable molecular marker for the diagnosis and therapeutic targeting of head-and-neck squamous cell carcinoma.
