Legumain Antibody (B-8) is a mouse monoclonal IgG1 kappa light chain antibody that detects Legumain protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-Legumain antibody (B-8) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Legumain, also known as LGMN, AEP (asparaginyl endopeptidase), or PRSC1, is a 433 amino acid protein primarily localized to lysosomes, where Legumain plays a crucial role in protein degradation and processing. Such lysosomal localization enables Legumain to participate in hydrolyzing asparaginyl and aspartyl bonds, essential for proper antigen presentation via MHC class II molecules in immune responses. Legumain is expressed ubiquitously, with particularly high levels in placenta, heart, and kidney, existing in both precursor and mature forms, with mature forms produced in dendritic cells. Legumain overexpression has been linked to solid tumor development, suggesting involvement in carcinogenesis. Multiple Legumain isoforms arise from alternative splicing, contributing to diverse functional roles in cellular processes.

Thank you for your support. I purchased Legumain antibodies at your company. I stained with immunofluorescent histochemistry, but I can't stain it well. I use 4% paraformaldehyde for tissue fixation, but is it better to fix it with methanol?

I am using Legumain (B-8): sc-133234 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Rated 5 out of 5 by from gave strong bandLegumain antibody can diluted 1:800 for Hela cell detection by WB.We used Hela cell as positive control,it's shows positive band at 50kda.I suggest use 5% BSA in TBS (without Tween) for blocking at RT for 1hour.This blocking condition is optimized ,no background.
