Lamin A/C Antibody (E-1) is a mouse monoclonal IgG1 kappa light chain antibody that detects Lamin A/C protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). anti-Lamin A/C antibody (E-1) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Lamin A/C plays a crucial role in maintaining the structural integrity of the nuclear envelope, which is essential for proper cellular function and organization. Lamin A/C (E-1) antibody targets proteins located in the inner nuclear membrane that provide mechanical support to the nucleus, thereby influencing gene expression and cell cycle regulation. Disruption of Lamin A/C can lead to various diseases, including muscular dystrophies and premature aging syndromes, highlighting Lamin A/C′s importance in cellular health. Lamin A interacts with several proteins involved in nuclear organization and chromatin dynamics, further underscoring Lamin A/C′s significance in cellular processes. Nuclear Lamin A is cleaved by Mch2, while Lamin C, a splice variant of Lamin A, differs only at the carboxy-terminus, sharing the first 566 amino acids with Lamin A.

Does this antibody recognize progerin?

Thank you for your question. Progerin has a C-terminal 50 amino acid deletion. As this antibody recognizes the N-terminus of Lamin, it should also be able to detect Progerin.

What retrieval would you suggest for the staining of paraffin sections?

Thank you for your question. You can find this information in our recommended protocol, which is available here: https://www.scbt.com/resources/protocols/immunoperoxidase-staining Our recommended method is heat treatment. Place slides in a container and cover with 10 mM sodium citrate buffer, pH 6.0 (sc-294091); or with 50 mM glycine-HCl buffer (sc-29096), pH 3.5, with 0.01% (w/v) EDTA (sc-29092). Heat at 95° C for 5 minutes. Top off with fresh buffer and heat at 95° C for 5 minutes (optimal incubation time may vary for each tissue type). Allow slides to cool in the buffer for approximately 20 minutes. Wash in deionized H2O three times for 2 minutes each. Aspirate excess liquid from slides.

I have seen that LMNA antibody (N-18, sc-6215) has been used in several publications related to IPSC-derived cardiomyocytes. Since this product has been discontinued, I would like to know the similarities or differences of this antibody and sc-6215.Thanks

Thank you for your question. Both antibodies are made against a peptide mapping at the N terminus of Lamin A/C of human origin (accession#P02545). sc-376248 is an excellent mouse monoclonal antibody that is recommended for the same applications.

I received a sample of lamin A/C (E-1) (sc-376248); what is the recommended secondary for this? I am using it to blot for mouse lamin.

Thank you for your question. This antibody is a mouse monoclonal IgG1 (kappa light chain). The recommended secondary is our m-IgGκ BP-HRP: sc-516102, mouse IgG kappa binding protein (m-IgGκ BP) conjugated to Horseradish Peroxidase (HRP). For other conjugates, please check on our website.

Secondary antibody was goat anti-mouse IgG1 AlexaFluor 488 (1:200). Staining was on sections previously stained with Laminin (also in green) and DAPI (blue) to identify myonuclei. This antibody provided beautiful staining with little effort!
