LAIR-1 Antibody (G-1) is a mouse monoclonal IgG2b antibody that detects leukocyte-associated Ig-like receptor-1 (LAIR-1) in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). LAIR-1 is a transmembrane glycoprotein predominantly expressed on human peripheral blood mononuclear leukocytes, playing a crucial role in immune regulation. LAIR-1 is located on the cell surface, where LAIR-1 functions as an inhibitory receptor that modulates immune responses by recruiting SHP-1, a phosphatase that negatively regulates cell signaling pathways. This interaction is vital for maintaining the balance of immune activation and inhibition, particularly in T lymphocytes, where LAIR-1 helps establish signaling thresholds essential for T cell receptor (TCR) signaling. The phosphorylation of LAIR-1 at specific tyrosine residues, such as Tyr 233 and Tyr 263, is critical for LAIR-1 function, as phosphorylation facilitates SHP-1 recruitment and influences downstream signaling events. Notably, LAIR-1 engagement on myelomonocytic leukemic cell lines has been shown to inhibit cell proliferation and induce programmed cell death, while cross-linking LAIR-1 on natural killer (NK) cells significantly dampens their cytotoxic activity.
