LAIR-1 Antibody (E-8) is a mouse monoclonal IgG2b antibody that detects leukocyte-associated Ig-like receptor-1 (LAIR-1) in mouse and rat samples through western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). LAIR-1 is a transmembrane glycoprotein predominantly expressed on human peripheral blood mononuclear leukocytes, playing a crucial role in immune regulation. LAIR-1 (E-8) antibody recognizes a protein located on the cell surface that functions as an inhibitory receptor, modulating immune responses by recruiting SHP-1, a phosphatase that negatively regulates cell signaling pathways. This interaction maintains T cell receptor signaling thresholds, essential for proper T cell activation and function. LAIR-1 occupancy on myelomonocytic leukemic cell lines inhibits cell proliferation and induces programmed cell death, highlighting LAIR-1′s potential as a therapeutic target in cancer. Cross-linking of LAIR-1 on natural killer cells significantly impairs cytotoxic activity, underscoring LAIR-1′s importance in regulating immune responses. Phosphorylation of LAIR-1 at Tyr 233 and Tyr 263 is mediated by Src family protein kinases, indicating a complex regulatory mechanism influencing immune signaling function.
