
JNK1/2 Antibody (E-5) is a mouse monoclonal IgG1 kappa light chain antibody that targets the c-Jun N-terminal kinases JNK1 and JNK2, which are critical in regulating cellular stress responses. JNK1/2 (E-5) monoclonal antibody was raised against amino acids 1-384 representing the full length of the human JNK1 p46 protein. Anti-JNK1/2 antibody (E-5) reacts with JNK1 and JNK2 p46 and p54 isoforms in human, mouse, and rat species, and can be used for western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). JNK proteins, derived from three genes, give rise to 10 isoforms through alternative mRNA splicing. The p46 isoforms include JNK1a1, JNK1b1, JNK2a1, JNK2b1, and JNK3a1, while the p54 isoforms include JNK1a2, JNK1b2, JNK2a2, JNK2b2, and JNK3b2. JNKs phosphorylate and enhance the transcriptional activity of c-Jun, coordinating cellular responses to stress and regulating cell growth and transformation. The human JNK1 gene (PRKM8, SAPK1, MAPK8) maps to chromosome 10q11.22 and shares 83% amino acid identity with JNK2. JNK1 is essential for the normal activation and differentiation of CD4+ helper T (TH) cells into TH1 and TH2 effector cells, with implications for immune response regulation. Moreover, JNKs are involved in several types of cancer, including breast, liver, and lung cancers, where dysregulation in this pathway promotes tumor cell survival, proliferation, and resistance to apoptosis. Capsaicin activates JNK1 and p38 in ras-transformed human breast epithelial cells, while nitrogen oxides (NOx) upregulate JNK1 alongside other signaling kinases such as MEKK1 and p38. In terms of post-translational modifications, JNK1 mediates the phosphorylation of Bcl-2, leading to the dissociation of Beclin-1, which triggers autophagy under stress conditions. This modification helps maintain cellular survival during nutrient deprivation but can also drive tumor progression when dysregulated.
