ISG15 Antibody (F-9) is a mouse monoclonal IgG1 kappa light chain antibody that detects ISG15 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). ISG15 monoclonal antibody (F-9) is raised against amino acids 1-150 mapping at the N-terminus of ISG15 of human origin. Anti-ISG15 antibody (F-9) is available in non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. ISG15 (F-9) antibody is composed of two ubiquitin-like domains, a structural feature crucial for conjugating to target proteins and regulating their stability, activity, and localization, thereby modulating key immune signaling pathways. The interferon-induced 15 kDa protein (ISG15) plays a crucial role in immune response by functioning as a ubiquitin-like modifier, conjugating to target proteins such as JAK1 and MAPK3/ERK1 through a unique enzymatic pathway distinct from ubiquitin. This modification regulates various cellular processes, including protein stability and signaling pathways, which influence immune cell activation and response. ISG15 exhibits specific chemotactic activity towards neutrophils, promoting activation and release of eosinophil chemotactic factors, thereby enhancing immune response. Additionally, ISG15 (F-9) monoclonal antibody is involved in paracrine, autocrine, and endocrine signaling mechanisms, notably inducing interferon-gamma secretion by monocytes and macrophages, essential for effective immune communication. Predominantly expressed in muscle, epithelial, neuronal, and lymphoid cells, ISG15′s cytoplasmic localization underscores importance in various cellular functions and interactions within the immune system.

-The datasheet specifies this is "a mouse monoclonal antibody raised against amino acids 1-150" but ISG15 is 165 aa. Is there more information on the epitope (not highlighted in the protein sequence tab)? -5% milk in TBS 0.1% Tween 4 C overnighincubation?

Thank you for your question. Yes, this information is correct, the antibody is raised against a recombinant protein corresponding to amino acids 1-150 of ISG15 of human origin. The exact epitope recognized by this monoclonal antibody has not been determined. We do recommend blocking in 5% nonfat milk in TBST (0.05% Tween-20) for one hour at room temperature, and incubating the primary antibody at the starting dilution of 1:200 in 5% nonfat milk + TBST for one hour at room temperature, or overnight at 4C.
