IRS-1 Antibody (H-7) is a mouse monoclonal IgG1 kappa light chain antibody that detects IRS-1 of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). IRS-1 (H-7) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The insulin receptor substrate-1 (IRS-1) plays a crucial role in insulin signaling as a major substrate of the insulin receptor, being phosphorylated in response to insulin, insulin-like growth factor 1 (IGF-1), and interleukin 4 (IL-4) stimulation. Phosphorylation occurs on serine, threonine, and tyrosine residues across various tissues, with casein kinase II being a key serine/threonine kinase that mediates a significant portion of insulin-stimulated phosphorylation of IRS-1. Notably, threonine 502 has been identified as the primary phosphorylation site for casein kinase II in rat IRS-1, while serine 99 serves as an additional phosphorylation site. Phosphorylation of IRS-1 by casein kinase II is an integral part of the intracellular insulin signaling cascade, influencing metabolic responses to insulin. Furthermore, IRS-1 contains three putative binding sites for the 14-3-3 protein, which are critical for insulin receptor interaction. Binding of 14-3-3 to IRS-1 is significantly enhanced following treatment with okadaic acid, a potent inhibitor of serine/threonine phosphatases, suggesting that 14-3-3 may modulate insulin sensitivity by disrupting IRS-1 and insulin receptor interaction.
