Involucrin Antibody (SY8) is a mouse monoclonal IgG1 antibody that detects involucrin in human and porcine samples through applications such as western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). Involucrin plays a crucial role as a precursor protein in the formation of the keratinocyte cornified envelope, which is essential for skin barrier function and protection against environmental stressors. Involucrin is initially located in the cytosol of keratinocytes and is later cross-linked to membrane proteins by transglutaminase during terminal differentiation. Anti-involucrin antibody (SY8) recognizes a protein that undergoes post-translational modifications, specifically the covalent cross-linking of glutamine residues to other envelope precursors via ε-(γ-glutamyl) lysine bonds. This process not only contributes to the structural integrity of the cornified envelope but also allows a single involucrin molecule to form multiple cross-links across various spatial planes, thereby enhancing the robustness of the skin barrier. Involucrin is specifically expressed in various cell types, including Chinese hamster ovarian cells, PtK2 rat kangaroo kidney cells, and rat epidermal keratinocytes, making involucrin monoclonal antibody (SY8) a valuable tool for studying keratinocyte differentiation and skin biology.
