Int-6 Antibody (H-5) is a mouse monoclonal IgG1 antibody that detects Int-6 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Int-6, also known as eIF3e or eukaryotic translation initiation factor 3, subunit 6, plays a crucial role in regulating translation and protein degradation by interacting with several key complexes, including the eukaryotic translation initiation factor 3 (eIF3), the proteasome regulatory lid, and the constitutive photomorphogenesis 9 signalosome. Int-6 (H-5) antibody recognizes a protein essential for initiating protein synthesis through assembly of 40S ribosomal subunits on mRNA, which is vital for cell growth and function. Notably, Int-6 gene is a site of integration for the mouse mammary tumor virus (MMTV) in murine tumors, highlighting Int-6′s potential role in cancer biology. Furthermore, studies have shown that reducing Int-6 expression in HeLa cells can lead to significant defects in mitosis, including issues with spindle formation, chromosome segregation, and cytokinesis, which are associated with the inhibition of cyclin B-Cdk1 kinase activity. This inhibition is attributed to a prolonged inhibitory phosphorylated state of Cdk1, underscoring Int-6′s importance in maintaining proper cell cycle progression and potential implications in tumorigenesis.
