Insulin Rβ Antibody (C-4) is a mouse monoclonal IgG2a kappa light chain antibody designed to target amino acids 941-1010 of the human insulin receptor β (INSR) subunit. anti-INSR β antibody (C-4) is an essential tool for researchers investigating insulin signaling pathways, effectively recognizing INSR β across multiple species, including mouse, rat, human, equine, canine, and porcine, and is suitable for various applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). The insulin receptor is a critical heterodimeric protein composed of α and β subunits, where the β subunit possesses intrinsic tyrosine kinase activity necessary for propagating insulin′s effects on glucose uptake and metabolism. Upon insulin binding, INSR undergoes autophosphorylation and recruits a cascade of interacting proteins, including insulin receptor substrates (IRS1 and IRS2), SHC (Src Homology 2 domain-containing), and the PI3K (phosphoinositide 3-kinase) complex, which play pivotal roles in downstream signaling pathways such as the MAPK/ERK and PI3K/Akt pathways. These interactions facilitate various cellular responses, including glucose transport, glycogen synthesis, and cell growth. INSR β monoclonal antibody (C-4) detection of phosphorylated forms of INSR β enables detailed studies of these interactions and activation status of the receptor, which is crucial for understanding molecular mechanisms underlying insulin resistance in type 2 diabetes and autoimmune destruction of insulin-producing cells in type 1 diabetes. Additionally, INSR β (C-4) antibody is available in multiple conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates, providing versatility for diverse experimental setups. What is the reason that this antibody recognises ~130 kDa protein, while other Santa Cruz antibodies vs. Insulin Receptor beta reveal a band around ~95 kDa?

The 130 kDa processed insulin Rβ as detected by insulin Rβ Antibody (C-4): sc-373975 is described in PMID 2187866 “Processed 130-kDa alpha and 95-kDa beta subunits were also observed and contained approximately 20% as much protein as the proreceptor on a molar basis.” Please contact technical service for more information.
