SEE ALSO...

HuR Antibody (6A97) is a mouse monoclonal IgG1 kappa light chain antibody that detects HuR in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-HuR antibody (6A97) is available in a non-conjugated form, specifically targeting HuR, which is a member of the Elav-like family of RNA-binding proteins. These proteins play a crucial role in post-transcriptional regulation of gene expression, particularly in mRNA stability and translation. HuR, also known as Human antigen R or ELAVL1, is ubiquitously expressed and is notably overexpressed in various tumors, highlighting its importance in cancer biology. HuR contains three RNA recognition motifs (RRM) that enable binding specifically to AU-rich elements (ARE) located in the 3′-untranslated regions of mRNA transcripts. This interaction is vital as ARE sites are responsible for targeting mRNA for rapid degradation, thus regulating the expression levels of genes involved in critical cellular processes such as growth and differentiation. By binding to these ARE sites, HuR inhibits mRNA degradation, leading to increased stability of the transcripts and ultimately influencing cellular responses. HuR shuttles between the nucleus and cytoplasm, a process regulated by a nuclear export signal that modulates accessibility to ARE sites, thereby playing a significant role in the post-transcriptional control of gene expression.

I am using HuR (6A97): sc-71290 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Asked by: jenniferc
We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
