HPV16 E7 Antibody (ED17) is a mouse monoclonal IgG1 kappa light chain antibody that detects the HPV16 E7 protein of HPV-16 origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-HPV16 E7 antibody (ED17) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. HPV16 E7 is a small zinc-binding phosphoprotein that plays a critical role in the regulation of the cell cycle by interacting with the retinoblastoma (pRb) tumor suppressor protein. This interaction leads to the disruption of pRb′s function, promoting uncontrolled cell proliferation, which is a hallmark of cancer. HPV16 E7′s ability to bind pRb is facilitated by the presence of the LXCXE motif, a sequence that is also found in other viral oncoproteins, such as the E1A protein of adenoviruses. Notably, high-risk HPV E7 proteins, including HPV16 E7, exhibit a significantly higher affinity for pRb compared to their low-risk counterparts, which underscores their role in oncogenesis. HPV16 E7 is predominantly localized in the nucleus, where HPV16 E7 exerts effects on cell cycle regulation, and HPV16 E7′s association with pRb not only promotes cell proliferation but also leads to the degradation of Rb family proteins, further enhancing oncogenic potential. Given the strong association of HPV16 and HPV18 with cervical, vaginal, and vulvar malignancies, HPV16 E7 monoclonal antibody (ED17) serves as a valuable tool for research into HPV-related cancers and the underlying mechanisms of viral oncogenesis.
