HPV16 E7 Antibody (NM2) is a mouse monoclonal IgG2a kappa light chain antibody that detects the E7 protein of HPV-16 origin by western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). HPV16 E7 (NM2) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The E7 protein is a small zinc-binding phosphoprotein that plays a crucial role in the regulation of the cell cycle by interacting with the retinoblastoma tumor suppressor protein (pRb). This interaction leads to the disruption of pRb′s function, promoting uncontrolled cell proliferation, which is a hallmark of cancer. The E7 protein′s ability to bind pRb is mediated by the LXCXE motif located within the CR2 homology region, which is also found in other viral oncoproteins, such as the E1A protein of adenoviruses. High-risk HPV E7 proteins, including HPV16 E7, exhibit a ten-fold greater affinity for pRb compared to their low-risk counterparts, thereby enhancing their oncogenic potential. The CR2 region contains a phosphorylation site for casein kinase II, which may further influence E7 protein′s activity and interactions. Given the strong association of HPV16 and HPV18 with cervical, vaginal, and vulvar malignancies, anti-HPV16 E7 antibody (NM2) serves as a valuable tool for research into HPV-related cancers and the underlying mechanisms of tumorigenesis.

I am interested in HPV16 E6 (C1P5)and HPV16 E7 (NM2) antibodies. I can see that you sell both antibodies conjugated with FITC, PE and different Alexa Flour. I was wondering if you know if one could use these antibodies in flow cytometry staining? Thanks

Thank you for your question. HPV16 E7 Antibody (NM2): sc-65711 is available conjugated to Alexa Fluor® 488, Alexa Fluor® 546, Alexa Fluor® 594 or Alexa Fluor® 647, which can be used in flow cytometry.

Can you recommend a positive control to use with HPV16 E7 Antibody (NM2): sc-65711 for Western blot?

We recommend Ca Ski whole cell lysate (sc-364360) to use as positive control as we have Western blot data showing specific detection of HPV16 E7 in this cell line.
