HPA1 Antibody (B-4) is a mouse monoclonal IgG2a antibody that detects heparanase 1 (HPA1) in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). HPA1 plays a crucial role in the degradation of heparan sulfate side chains of heparan sulfate proteoglycans in the extracellular matrix, which is vital for various physiological and pathological processes, including tumor metastasis and inflammation. By dismantling the subendothelial basal membrane, HPA1 facilitates the extravasation of blood-borne tumor cells and inflammatory leukocytes, thereby promoting cancer progression and angiogenesis. HPA1′s ability to release growth factors and cytokines upon degradation further stimulates cell proliferation and chemotaxis, highlighting HPA1′s importance in tumor vascularization. Additionally, fibroblasts can endocytose extracellular HPA1, leading to accumulation in the cytoplasm, which may influence cellular responses to the tumor microenvironment. The gene encoding human HPA1 is located on chromosome 4q21.3, underscoring HPA1′s significance in human health and disease. Anti-HPA1 antibody (B-4) is an essential tool for researchers investigating the roles of heparanase in cancer biology and inflammation.
